I agree that tadalafil and sildenafil likely have similar longevity effects and there is research to support the use of either for longevity impact. On that note sildenafil likely has a similar side-effect profile to tadalafil, so I’m not seeing much difference there. While you argue tadalafil’s longer half life means longer exposure to the side effects, it could also be argued that it means longer exposure to the vascular and other longevity benefits. In the case of rapamycin, longer exposure to the drug is believed to activate mTorc2, which some hypothesize is responsible for rapamycin side effects, so we’d naturally want to pulse our exposure. However, I see no evidence for a similar mechanism for PDE-5 inhibitors. Anyhow, it’s an interesting thought experiment, but I’m not imagining the outcome would be significant.

That’s the ED dose, good for a dirty weekend. The research for BPH and vascular impacts seems to land on 2.5 to 5 mg daily, which is also helpful for ED. Tolerance can develop so taking a break now and again is recommended.

Ha! This paper does report a doubling of muscle protein synthesis:

This is with a week of daily sidenafil dosing and no training! However, this study of only 11 subjects is a long way from “scientifically proving . . . . this ED drug could catapult your gains.” As the authors note “the short‐term nature of the study prevents conclusions regarding possible effects of chronic sildenafil therapy on muscle mass or mitochondrial biogenesis. Such endpoints, as well as the possibility of reduced effectiveness with chronic use (tachyphylaxis), will need to be addressed in longer‐term studies.” As far as I can find, no such long term studies have been done in the 11 years since this study was published.

Do the GERD-like symptoms dissipate with chronic use? I’ve tried sildenafil a handful of times, but the GI disturbances have kept me from running it more frequently.

The GERD never went away for me, which is why I eventually had to give up. Even 2.5mg was more trouble than it was worth. Stuffy nose went away but the feeling of needing to burp because of air being trapped in my esophagus all the time did not.

I forgot to mention in my previous message that tadalafil also caused near vision impairment for me. As I understand, tadalafil is associated with glaucoma in some long-term follow-up studies. Probably a negligible risk, but good to keep in mind if symptoms appear. Glaucoma can lead to blindness.

I’m sorry to hear this. What were your initial symptoms of this, if I can ask? Bloodshot eyes appear to be mentioned quite often on for example on Reddit, even on low doses. (Not the most reliable source, I realise). As well as feelings of some eye pressure.

I’m not sure if ‘visual impairment’ is the right term, but I couldn’t read properly at times, and now I can see normally again. My eyes weren’t very red.

I definitely got the bloodshot eyes from it. That never went away.

2.5mg daily Tadalafil for past few weeks. No negative side effects. Perhaps slight decrease in BP which is desirable for me.

I normally take 20mg once a week during the day. Anyway after reading I decided to half the pill and take it last night. Almost within 2 hours my nose is completely blocked. Never had this issue before. What is mechanism that caused it?

That’s what we are all trying to figure out. Right now, it looks like the evidence is slightly pushing towards the direction of Sildenafil, but the case is far from closed.

I agree. IMO: Because, oddly, some people feel adverse side effects from one but not the other, I would take whichever one I tolerated best.
As I have previously posted, I prefer sildenafil because of its shorter half-life, which makes dosing more straightforward. FWIW: I have been taking sildenafil, 25mg daily, for quite some time without any subjective side effects.

Also, sildenafil is the drug that demonstrated longevity benefits. We only accept that tadalafil does the same thing because of its similarity to sildenafil.
