Hi, I’m curious if there’s anyone in their 20s/30s taking rapamycin here and at what dose?

I’m 31 and I’ve been taking 3-4mg/week for the past 2 months. I did a 2 week washout and did another blood test. My lipids went up about 20% which is in line with other posts I’ve seen here. I’ve also lost 10 lbs (150lb → 139lb @ 5’10") and likely a few more since I last checked. I’ve lost enough to the point where I need new pants. There was no change in my usual diet and exercise. Rapamycin seems to promote lipolysis so it makes sense, but I wasn’t expecting that big of an effect.

Total cholesterol: 212 → 244
Triglycerides: 104 → 127
HDL: 61 → 64
VLDL: 18 → 22
LDL: 133 → 158
LDL/HDL ratio: 2.2 → 2.5
ApoB: 113 (test 2 months ago didn’t measure ApoB)

I chatted with someone from the clinical team at GetHealthspan and they suggested I try doing 3 months a year due to my relatively young age. I still have ~18 1mg tablets and they recommended lowering the dose and frequency to 2mg every 2 weeks since it could’ve been accumulating in my body at my original dosing schedule. I’m thinking of finishing the bottle @ 2mg/2weeks and waiting until next year to resume.

For anyone in their 20s/30s, what dosing schedule are you doing and have you seen any metabolic side effects too?

So I am 30 and take 12mg every two weeks, I got one test before starting, one test while taking and another after a one month washout. No substantial effects on triglycerides (although some tests were fasting and some not). LDL had a small increase from low 40s to low 50s and is now back to low 40s after a month off. HDL was low 30s before and during but has now gone up to high 30s after a month’s washout. Overall a very minimal effect for me.

This thread has more discussion on it. Regarding time off I think it is a good idea especially for younger people. My plan is to take two 8 week breaks per year so roughly 8 months on 4 off. I also donate blood twice per year so the 2 weeks before and 6 weeks after blood donation I am off rapamycin.

I’m 31 y/o and on my second cycle of Rapamycin. I’m currently taking 5mg weekly - as a 170lb male.

Last year, during my first time on rapa, my non-HDL increased by 50% and my dr immediately prescribed statins.

This time around, I’ve been taking 5mg rosuvastatin twice weekly.

My plan is to cycle on and off rapa quarterly - using lipid lowering agents while on cycle.

I have never experienced weight loss on rapamycin. I lift 5 days per week + run 30 miles per week + HIIT once per week. I have seen no detrimental impact on muscle gain - so no weight loss. Feel great honestly.

I should clarify, my weight loss seemed to be mostly, if not exclusively, fat around my waist area. I did not appear to lose any muscle mass as far as I can tell.

If your ApoB and LDL are high for any reason, you should consider Bempedoic Acid, Ezetemibe and/or a statin before Rapamycin (or all 3 like I do). CVD has a higher chance of taking you out before old age does.

My LDL (and ApoB) are now 48, down from 120 after Rapa and 67 before Rapa. My LDL and ApoB move in almost perfect lockstep for some reason.
