How were your concentrations/doses selected? 50 uM seems extraordinarily high for any drug. While SGLT may not be its mechanism of longevity enhancement, empagliflozin would provide 80% inhibition of SGLT-2 at about 12 nM. I would be concerned 50000 nM ( = 50 uM) would have nonspecific toxicities.
When people do in vitro testing, 10 uM is often used as a default real high concentration to see if anything happens. But unless you’re looking at the liver or a few other edge cases, biologically relevant concentrations of drugs are usually below 1 uM.

Can you please ask them? (on Twitter or by email) I don’t know enough about this issue but they said once that before testing something they look at a dose response with 3 different doses.
Sure. I’ll post if/when I get a response to my email.

I asked them and they said default they test at the maximum concentration that dissolves since worms have thick skin and they want to make sure it has an affect. Then they can try lower doses later. It doesnt seem they think too high doses is a big deal. For berberine or tumeric they cant get enough to dissolve to match other papers

Ok, this is funny to me. @AnUser accused me of believing in magic crystals and shungite:
https://www.amazon.com/Elite-Shungite-Stone-Purification-Mineral/dp/B071ZZ93FK/
Shungite is a magic crystal that is said to purify water or if you wear it around your neck it will keep away bad vibes. It is almost pure carbon and the form is mostly fullerines or buckyballs. So I wanted to have shungite tested in the Million molecule challenge. Emailed them and got this back:
Thank you for your interest in the Million Molecule Challenge and Ora Biomedical!
** We have not tested shungite. For safety and liability, we must source compounds from reputable research grade chemical suppliers.**
** That said, in my quick literature search, fullerene may be a suitable replacement. That is offered by chemical suppliers. To run that test, we would need to charge for compound purchase and shipping. An estimate for a single dose test would be around $400.**
** Let me know if you are interested and I can draft a custom contract.**
So the price is a little higher than I wanted, also not testing shungite (obviously magic for some reason) but fullerines. I have no idea how much to use or whether to mix it in the feed or just use it for some form of bedding to ward off bad vibes.
Does anybody have a good idea here?

Yeah, I knew about c-60 and couldn’t find it on the list either. I’m surprised they didn’t have it on there. Sarah Westall sells it on her program all the time. Supposed to cure all kinds of stuff. LOL.

Yes, it is a little pricey even if you make your own.
Frome SES Research:
"SINGLE BOTTLE ESS60 AVOCADO OIL
ESS60 - Carbon 60, 99.99% in Avocado Oil Extra Virgin, Organic, 120ml" $79.95"
From “https://www.sesres.com/carbon-60-supplements/c60-in-avocado-oil/”
I agree with @Bicept, that avocado oil is your best bet to avoid digestive tract problems.
I tried C60 in olive oil several years ago but felt no subjective benefits, I didn’t notice any effect on my blood work so I didn’t continue taking it.
I’m sorry but there is little proof that this does anything in humans.
FWIW: Here are two YouTube videos showing step-by-step how to make your own C60 Olive Oil.

Interesting to see that Curcurma, Apigenin and Pterostilbene quite significantly reduced lifespan, with these worms at least.

I didn’t see the dosing. Give that these are all HDAC inhibitors (which IMO are harmful in high doses, but beneficial in small doses see Queen Bees/Royal Jelly and beta Hydroxybutyrate during keto) it worries me if the worms respond badly to HDACi whilst other species respond well to that.
HDACi can cause apoptosis through ATP depletion in large quantities and would cause ATP shortage if not high enough to kill.
I suppose it comes down to a dosage level really.

I agree John. I think they should test at 3 levels for everything. Otherwise you’re just shooting blind. The dosage is very important.

Are people allowed to choose the dosage, or what’s going on?

I answered this a few posts above. You are free to email them to setup a custom experiment however.
Just remember that things that are bad for worms are not necessarily bad for mammals. Likewise things that are good for worms may or may not be good for mammals.
This experiment is to identify things that are good for worms and then try them out on mammals to see if they work.
This process isn’t perfect.

It appears one of mine was tested and one i think the did a different chemical than i asked for. I hope they have good procedure to make sure they are testing the chemical they say they are at least
Edit: i havent gotten any emails back so it may be someone with a similar name.

My results arrived and they all failed: atorvastatin, bimatoprost, flecainide acetate, hydrochlorothiazide, indapamide, isradipine (Dynacirc), lithium carbonate, lithium citrate, naphazoline HCl, sitagliptin, and terazosin HCl.
I’m waiting for the results of the other compounds I sponsored:
- Semaglutide
- Selegiline
- Obicetrapib
- Urolithin A
- Telmisartan + Amlodipine Besylate
- Amlodipine Besylate
- Lercanidipine
- Lacidipine
- Felodipine
- Nitrendipine
- Carvedilol

Thanks for sponsoring these. It’s a bummer that so many are failing though.
