I do understand it quite well, but I do also understand biases behind it and also why for some mechanistically sound concepts there is almost impossible to find funds to do further study evaluations.

I am doing it to kill senescent cells only- nothing else. But this form with all the scary post is making me rethink my decision to continue although I consider myself healthy! But I can find 20 papers on Rapa that have scary side effects too!

I appreciate all the concern about the side effects but seriously on this form? IMO most all taking Rapa are taking a risk, you really never know what lurking in our bodies and DNA.

So why are you comparing a mechanistic study with clinical trials then? As if no one has more information about a drugs efficacy based on a clinical trial compared to mechanistic studies?

Rapamycin doesnā€™t have a post-market warnings from the FDA as I showed in a thread about this.

I appreciate being made aware of any apparent potential dangers of taking Azithromycin. However we need to keep things in context. The DAV therapy that people are trying here uses a total of 10 Azithromycin 250mg doses over a period of five weeks. That is nowhere close to the standard antibiotics regimen where any potential for elevated risk has been observed. In fact the trial cited that was aborted had been using ā€œlong termā€ Azithromycin therapy. It would appear yet again that it is probably the dose that makes the poison.

The dose with increase in cancer relapse and death was 250 mg azithromycin 3 times weekly. The translated article protocol does 250 mg two times a week. Not sure why you call that subclinical dose, if itā€™s only 34% lower than the dose showed to increase cancer relapse and death from long term use in a clinical trial.

It calls into question the ā€˜efficacyā€™ of this therapy if a 50% higher dose for one of the drugs increases cancer relapse and death in a clinical trial. Nothing extraordinary as itā€™s typical for mechanistic studies to fail or even be harmful as a therapy is studied further.

Yes, there might be people with CVD here so for them itā€™s good to know and not an automatic ā€˜no brainerā€™ as it apparently is for some.

See my post above. The patients took doxycycline for thirteen months.

243 patients were to receive > > azithromycin (250 mg) 3 times weekly for 2 years; 237 patients were to receive placebo for two > years, starting at the time of conditioning regimen. The immunomodulating effects of > azithromycin therapy were evaluated when used for the long term prevention of BOS.

Results: The ALLOZITHRO study treatments (azithromycin/placebo) were terminated on the
> 26th December 2016 i.e. at thirteen months after completing recruitment

Hmm. I was using some hyperbole in my statement. Hard to read tone in a blog post. btw - if you have read any of the science on human relationships and friendship you will see pretty conclusive evidence that giving advice (i.e. being officious) is always detrimental. It is related to horizontal and vertical relationships, humans work best with horizontal relationships - aka we are all the same, no one is better than anyone else.

I did read the research on doxy quite extensively because I had been taking it weekly with my rapa dose to prevent rogue bacteria from inhabiting my Birmingham hip device. Vitamin C is benign and most of it is peeā€™d out. Azithromycin is the wildcard and I have taken it before without issues. In fact as I have stated before I ā€˜feelā€™ better, have healthier hair and experience some of the claims in the research study used to kick off this thread. So risk reward to try this for 5 weeks where the Z is only every 3 days seems quite low to me. Also, I have joint issues, arthritis and there is some evidence that bacteria cause it. I didnā€™t explain myself for you @AnUser, but for others thinking about trying it.

As I have stated before, the only change I feel is less knee joint pain.

My modified DAV Therapy

Taking :

250mg of azithromycin once ever 72 hours

100mg of doxycycline every 8 hours {I did miss o few times the 8hour window, just moved the dosing forward]

2.4g{yes grams ] of Vitamin C, two types approximately 4 hours after taking either the azithromycin or the doxycycline

Have not had any side effects, been on this dosing since September 25, 2023 will be on this till October 31,2023 a total of 36 days.

Not taking rapamycin during my DAV Therapy dosing.

What would be common in health magazines, for example.

Itā€™s probably snake oil.

And it is being tried by several member of this forum.

Iā€™m really interested in trying this protocol as well, and have been trying to keep up with the members who are currently doing it, and whether they are following the ā€˜standard protocolā€™, whether they paused rapa use, their end date, and any other salient/important details. And later, of course, their perceived subjective as well as any objective test results.

Std protocol:
ā€œFor 5 weeks, azithromycin was administered at 250 mg twice weekly, doxycycline at 100 mg twice daily, and vitamin C at 500 mg once dailyā€

Here are the folks I know about from following this thread; are there others who would like to make themselves known?

KFISH: std protocol - rapa/all supplements paused - ended 11/1/23. Finished ā€œDAVā€ ā€˜reported no side effects, blood test came out perfect same as usual there was no changes to any longevity blood markers.ā€™
