oops, typo, meant glycosylation, not glycation.

Regarding knee (or any other pain), a counterpoint to medication (to heal damage or inflammation) is the idea that the brain uses pain to keep you from doing activities it feels are unsafe. The therapy is to teach the brain such movements are safe, and then the pain will go away. I’ve attached an excellent overview from Z-Health. I’m starting to explore this area now. Z-Health has many free videos on YouTube for specific therapies.

I do these and VMO squats as part of Bryan Johnson’s blueprint plan, they do help your knees.

Bryan Johnson has a new rapa dose schedule. It seems that he will re-up before zero based on 13mg/6mg weekly alteration. In the transplant patients cohort, doses are daily but higher, and yet benefits are seen based on retrospective studies.
A few experts have stated daily low-dose (1 or 2mg) or weekly-dose (6 or 13mg) benefit scenarios are probably 50 to 50. I wonder why has no one tried the daily low-dose regimen to see if indeed is no difference or perhaps better?

He does not seem to be concerned about science.

@Jonas, I’ve considered 0.5mg (I’m only 105 pounds) daily for one week on and one week off like I heard in one of @Krister_Kauppi ’s interviews of a Buck Institute scientist. I’m just waiting to reorder because I’m certain 1mg daily would be too much for me.

I started a low dose, 0.5mg, daily besides Sat and Sunday. Total per week is 2.5 mg. Still don’t feel any difference.

@LaraPo what was your previous dose?

I tried different doses as I had a kidney transplant 13 years ago. The prescribed dose was 1mg every day without any breaks. I was on that dose for a few years. Then I changed to 1 md/day for 4 days followed by a 12 -14 day break. Now I’m trying 0.5mg for 5 days followed by a 2 day break. My goal is to completely eliminate side effects while maintaining all healthy blood markets.

what side effects you have so far on previous dose of 1mg?

I didn’t have any side effects on 1mg for 4 days. The reason I switched to 0.5 mg for 5 days is that I want to reduce break time between doses bc it was suggested (by M Blagosklonny) that mTOR may get too active in the result of a longer break.

I have tried the 13 mg/6 mg protocol before. Euphoric fatigue was greater with the greater dose. What I found beneficial was an extra week to flush out any remaining Rapa after the 6 mg week. This is all based on feeling though.

I’m taking Rapa for psoriasis as well. So far so good. Will post my stack and results in a few weeks. If you have psoriasis, you definitely have an over-active immune system. It’s an auto-immune disease, and Rapa is an immune suppressant. Best of luck!

I’m debating whether or not 8g of daily taurine might be advisable for those of us who are larger.

I take 1g of taurine a day. As I see it
a) You get material amounts of taurine in diet
b) Some is endogenously generated.
Hence it is an interesting question as to whether there is a optimal amount to take for any one person. I would think there probably is, but also I would think it varies from person to person.
It strikes me that having some indication of taurine levels would be a good idea. Does anyone know to what extent say serum taurine levels are indicative of cellular levels?

I’m taking approximately (powder not measured carefully) 8g/day split between AM and PM. I haven’t noticed anything in general but do notice a boost in energy (feeling) and power (more reps) when taken before weight lifting.

Welcome @cara2023 I just noticed this post, and am so happy to meet a fellow “Patient”! Yeah, I have had eczema all my life on and off, doctors keep pushing prednisone cream which is mostly bad for you. Then I developed psoriasis (lol) after starting Dupixent. My eczema and psoriasis “competed” on different days. Anyhow, I discovered a small trial on psoriasis with Rapa so here I am! What dose regimen you are on? I am on a daily 1mg x 5 then I take a day or two off. I do better on the days that I am on rapa and I have not had many side effects that I noticed. Efficacy of sirolimus (rapamycin) administered concomitantly with a subtherapeutic dose of cyclosporin in the treatment of severe psoriasis: a randomized controlled trial - PubMed
