@adriank if you are relying on creatinine for your eGFR calc, you might try Cystatin-c instead to avoid any confounding with creatine supplementation or high muscle mass.

honestly i just get a lab testā€¦ they tell me the scoreā€¦ I have no idea whyā€¦ but next week i will see if it relates to the time i have my rapamycin dose and after a week get another test

Every time I take Rapa I have full exhaustion the following day. I was taking 4MG a week for a month but had to take a break as I was losing 24-36 hours each week from utter exhaustion. Like naps 2 hours after waking and again at 2pm

I took a break for a month but took 2mg on the 3rd. Same response but slightly less duration.

Is this a normal side effect and if so does one get used to the Rapa and does this side effect lesson.

This is something that many report. I have been taking a higher dose for almost 3 years.
Yes, the next day is a waste, as far as exercise energy goes, but I go to the gym anyway.
It has never made me sleepy at any dose.

Hey Marshall,
I can concur. I take my rapamycin at night, usually about 11 PM. And, when I wake in the morning at 7AM, Iā€™m really disoriented and foggy, (like a hangover). Clouded mind until about noon. Then, slower rest of dayā€¦ no gym workout.

Next dayā€¦ back to normal.

@marswalker Your experience mirrored (but not as bad as you described) my own until this last dose yesterday. Today I woke feeling the rapa but it left within a few minutes and I feel great today. And zero muscle soreness. Itā€™s very strange

Many things may have changed but the only thing I did for the first time intentionally was to do a moderate bike ride for 1.5 hours starting an hour after I took Rapa on an empty stomach (4mg with 3 grapefruit preceding the dose by 6/3/1 hoursā€¦much is different from my normal as a result of a last minute change of plan).

I will definitely repeat this protocol.

I am beginning month four after building up from 2mg to 6mg and am experiencing the same thing. I take my Rapa after a morning walk and drink black coffee and a bunch of water. I am wiped for the rest of the day which is fine because itā€™s my rest day. But, I wake the next morning completely groggy and the second day still feel tired, by the third day all energy is back. I canā€™t stand feeling tired for two days out of a week. I am going to try Agetronā€™s recommendation and try taking it the night before. Very similar to you, 60 yrs old 195lbs in decent shape.

Base on what you say isnā€™t just after dinner the best time? That way when we are ready to sleep it will coincide with the fatigueā€¦ I usually feel something around the 1.5 to 3 hour mark. After that depending on whether I want to dose up on the taurine or not. Usually I donā€™t.

Yes after dinnerā€¦ I work out in gym at 8 pm home by 10 pmā€¦ cook up a steakā€¦ potatoā€¦ milk. Take my rapamycin at 11 pm and crash.

Same meal for the past seven years after gym. Saturated fats are not bad for me

I can eat like that everyday minus the milk but that is a very late dinner. I usually try to have dinner by 6pm :wink: I am not a fan of asparagus, but there are lots of other vegetables I would add.

That is a delicious looking dinner! Mine is Pesco-vegetarian and doesnā€™t look nearly as delicious!!!

I donā€™t know if itā€™s better for.healthspan, but a dinner like that definitely improves the quality of life!!!

Weā€™re all different genetics and phenotypes. One of my closest friends cannot eat meatā€¦ doesnā€™t sit well. Heā€™s a pretty much absolute vegetarian, but if his wife tries his diet, she gets sick and has to have meat most meals.

So again, to each their own. All over the world, every place has a diet of that regionā€¦ which likely would be unhealthy for someone, not from that locality.

My backgroundā€¦very Neanderthal (grey eyesā€¦ natural red hair tint), my parents are Scottish and Czech.

Meatā€¦milk and potatoes :potato:. Your body will tell you what diet is best. Your gut will feel right and body will respond for you.

Love this meal including the milk part (I like to have a glass of milk with my meal once in a while). I would leave potato out even though I love potato, but for some weird reason I have been able to convince myself potatoes are bad and have almost stopped eating them altogether. I think iā€™ll need to revisit them as the cravings are still there LOL.

I rather die than skip on food I like lol. I am happy to omit certain foods but eating nice food is a priority in life.

@adriank Itā€™s ok to eat nice food. Just take your Rapamycin, Bempedoic Acid, and Acarbose to nullify the bad effects.

And also try to choose the healthiest nice foods. You wonā€™t live forever, but life will feel better.

Actually about Bempedoic Acid, is it really necessary to lower cholestrol? My cholestrol is very low for some strange reason. I thought we need higher cholestrol to live longer? (something I read recently). Plus I love my BBQ and fried foods. If I stop my chocolates for 3 months my LDL will drop.

BTW, what is CR?

According to your records, your latest LDL would be 96 in American terms. Anything above 50 is building plaque. Iā€™d be taking Bempedoic Acid and it should take you down to 50. I was at 120 and am now at 65. Itā€™s still building plaque but at a slower rate. Iā€™m going to try Atorvastatin to see if I can get it lower, but Iā€™m afraid I wonā€™t be able to tolerate it.

CR is Caloric Restriction.

As an aside, My LDL was at 67 before I started taking Rapamycin, so Bempedoic Acid + Ezetemibe is countering the hyperlipidemia from Rapamycin.

Started rapamycin on June 3 (5 weeks ago) as 1mg/wk x 2-3 wks, then 2mg/wk x 2 wks, and today 3mg.
PVCs: I noticed increased PVCs (extra heart beats) initially but those have disappeared.
Defecation Reflex: Also noticed (well, struck with) intense defecation urgency the day after a dose. That has improved-- I donā€™t have to use the park Porta-Potty when exercising, but can wait until I am home.
Exercise Regimen: I have been on the same exercise regimen for years: I arrive at a local dam at 6:30am and do 9000 stair steps over 1.5 hrs. The steps have an 8" rise x 111 steps up. I think I feel better, even on the day-of dosing and the following day than I did before rapa. My stairs are followed by push-ups. I go to exhaustion and could do 45-50 before rapa and that has decreased by 10%, again to exhaustion but we have had a heat wave which means more cardiac output goes to the skin for cooling than when it was cooler. I sweat horribly and drink up to 1/2 gallon of water while doing the steps in this weather.
Infections: Other than a brief episode of subclinical shingles (my diagnosis) initially, I have not had any infections, not even a folliculitis (pencil eraser-sized skin infection at the site of a hair root) common in those who exercise and sweat lots.
Blood sugars: I use a continuous glucose monitor (Dexcom G7), which I calibrate with a glucose meter. I do not eat bread, pasta, anything with added sugars, cereals, ice cream, etc. My fasting and postprandial glucose rose by 10% each based on lots of prior data. Today I am receiving from India remogliflozin (an SGLT2 inhibitor) that is inexpensive and efficacious and not sold in the US.
Plan: Continue to slowly increase dose, probably to 6mg.
