Yes, an excellent presentation on The Proof podcast. I knew about the ApoE connection to dementia but this illuminated how this apolipoprotein ties in with ApoB and ApoA1 and also the relationship between brain cholesterol and peripheral cholesterol. Tom Dayspring was at his best and it was a lively discussion. Fascinating subject.

Typically, individuals with early-stage Alzheimer’s disease perform progressively worse on standard neuropsychological tests, losing a few points every year. But in Chan’s study, participants’ test scores have remained stable. Brain volumes have shown no signs of shrinking and levels of a disease-associated protein in the blood have even trended downwards. “These patients have conquered the test of time,” says Chan, who presented the findings at an international conference on neurodegenerative diseases

This article briefly summarizes the role of magnesium, an essential mineral involved in numerous enzymatic reactions and critical for neuronal bioactivity, in the context of neuroinflammation and cognitive decline. The potential neuroprotective effects of magnesium, including the mechanisms of neuroprotection by magnesium through maintaining neuronal ion homeostasis, reducing inflammation, and preventing excitotoxicity, are also described. Additionally, we discuss the impact of inadequate magnesium on neuroinflammation and its potential as a therapeutic agent for attenuating cognitive decline to improve neurodegenerative conditions.

Buntanetap is a drug under investigation for AD and PD. It inhibits the production of neurotoxic proteins and has been shown to improve cognition in early AD patients (NCT05686044) and halt cognitive decline in PD patients (NCT05357989). Interestingly, last month, the company behind it filed three patents for combination therapies with dulaglutide and sildenafil @DrFraser!

Recent preclinical studies have demonstrated that when buntanetap is combined with a GLP-1 agonist (such as Trulicity) or PDE5 inhibitors (such as Viagra), cognitive function in Alzheimer’s mouse models improves to levels beyond those observed in healthy controls. With these compelling results demonstrating strong potential for cognition-enhancing combinations, Annovis further strengthens its intellectual property portfolio through the filing of the following patents:

  • Buntanetap + Trulicity (dulaglutide, a GLP-1 agonist).
  • Buntanetap + Viagra (sildenafil, a PDE5 inhibitor).
  • Buntanetap + Trulicity + Viagra.

We don’t have the preclinical data (and it’s in mice…) but it’s noteworthy that they chose dulaglutide (instead of exenatide, liraglutide, lixisenatide or semaglutide) and sildenafil (instead of tadalafil).

That being said, their stock is down 51% YTD so it might just be a desperate move to attract attention…

Semaglutide appears to be protective after just a few weeks. So it’s most likely confounding BS. Unfortunately.
