Hi Dr Ross, thank you for that very informative YouTube presentation. I am an 80 year old women who has had to watch my glucose level, in doing that I have lost weight from 140lb to 125lb. 2 questions, does the dose of Rapamycin alter according to one’s age? And secondly, , when I tried Rapamycin 8 months ago, my glucose level spiked, will the glucose level return to the non spiked level a day or 2 after consuming Rapa? Thank you

Question #1. To my knowledge, no one has studied if rapamycin doses should be adjusted for age. It is a good question, but we still have much to learn. A similar question could be asked regarding body weight; should a person weighing 250 pounds take a larger dose than a person weighing 150 pounds. We just don’t know yet. However, taking 5 or 6 mg once weekly seems to be safe and effective for most people…but we don’t know if that is the optimal dose or the optimal plasma level of rapamycin/sirolimus following a dose.
Question #2. A few people, but not many, have reported increases in blood glucose after taking rapamycin. With this question, we also don’t have studies to provide definite answers. To my knowledge, people reporting elevations of blood glucose have not tested to determine how quickly levels return to normal following ingestion of a dose.
Here is one human clinical trial that does provide some helpful information:
A randomized control trial to establish the feasibility and safety of rapamycin treatment in an older human cohort: Immunological, physical performance, and cognitive effects
The authors state the following: Oral glucose tolerance tests revealed no RAPA-induced change in blood glucose concentration, insulin secretion, and insulin sensitivity. Thus, based on the results of our pilot study, it appears that short-term RAPA treatment can be used safely in older persons who are otherwise healthy.

Interesting. Could you please give more details here. Was this your blood level at 20 hours after taking rapamycin? What dose did you take when you got these levels and what is your body weight, if you don’t mind sharing?
