Its been a few years since I researched this, but transcutol is a very safe product from what I’ve read. Its regularly used in oral medication formulations. As a toothpaste or flossing paste, I use a small amount - under 5% of the total formulation, and of course most of it I spit out after brushing or flossing.

@Rapadmin Thanks so much for this. i am late to the party and just finding out about Rapa for teeth and gums! I have done quite a bit of reading about it and understand that rapa doesnt go into the bloodstream when used with a body lotion or toothpaste. Can i ask what the benefit would be of also taking a rapa tablet orally along with the toothpaste? Or is there one? I read about how it turns off the mTor so shouldnt be used daily, but instead maybe weekly only. Wanted to know if taking a weekly 1-5 mg. tablet orally would help with the tooth and gum also or would i experience something more I havent learned about yet. Thank you again for posting this! Have been looking for exactly this for awhile now!
The short answer is we don’t know for sure (in humans). In mice it was the oral rapamycin that triggered the interest in seeing if the topical rapamycin would be helpful for dental applications. So - in theory it could be an additive benefit to combine the topical/oral toothpaste with the orally taken rapamycin tablet; but we really have no human data yet.
In terms of schedule - we don’t really know and have no good data. In the topical skin cream formulation study they used rapamycin cream on the skin every day, without problems. Rapamycin May Slow Skin Aging (Drexel U. Study)
So, I’ve just used my rapamycin toothpaste as I use my regular toothpaste.

Thank you so much for the fast reply! May i ask if you have noticed and remineralization on your teeth or, more importantly at least to me, have you experienced any regrowth of your gums? I see this thread started a couple of years back so would be interested to learn any results. Thanks again!

@RapAdmin Absolutely love this and thanks for posting! Question tho…Can you weigh in on whether or not powdered rapa could be used instead of tabets that need to be crushed? It seems like the powder would be alot simpler to use. Thanks!
Yes, absolutely. Either way is fine. I do think powder is easier, but most people already have the tablets on hand.

In your ingredients list you posted Zydus Sirolimus. Is Zydus the place you ordered from? I am new to ordering overseas so am trying to learn as much as i can about it. And is Zydus a place you trust and have had good success with? Thanks much!

It depends where you live to some extent. Rules on importing molecules vary by country. Zydus wont sell directly you need an online retailer. See the other threads for details.

So grateful for this post! We found a good source for our rapamycin and are waiting for its arrival so now we are focused on gathering the other necessary ingredients for our toothpaste. Can i ask did you add the transcutol as a way to enhance the rapa absorption? After reading the transcutol safety background article in your post (thanks for that BTW:)

No - you need something to help the rapamycin get absorbed by the skin & tissue in the mouth - like Transcutol.
sirolimus is an extremely hydrophobic compound and sensitive to temperature, light and pH. Its poor aqueous solubility results in a slow dissolution rate and consequential low oral bioavailability.

Yes, you definitely want the transcutol for increased absorption (IMHO), or some similar agent. I think I used LabAlley.com for getting the transcutol, but its available from many places. All the instructions are very similar to the Skin Cream DIY instructions, we just use toothpaste to replace the skin cream: DIY Rapamycin skin cream
